Uma pesquisa da Universitat Politècnica de València (UPV) na Espanha desenvolveu um anti-inflamatório que pode ser uma alternativa ao uso de corticosteroides no tratamento de COVID-19 e doenças inflamatórias crônicas. O novo medicamento foi desenvolvido a partir de ingredientes de uma planta medicinal originária do Sudeste Asiático.

Tratamento alternativo
O tratamento com corticosteroides – tipo de medicamento anti-inflamatório – começou a ser utilizado precocemente em indivíduos infectados com SARS-CoV-2, logo no início da pandemia. O objetivo era tratar uma condição chamada “tempestade de citocinas”. Ela ocorre quando o sistema imunológico reage de forma muito intensa à infecção.
Os corticosteroides foram usados para tentar controlar essa resposta inflamatória excessiva. No entanto, pesquisas posteriores revelaram que a administração do medicamento em pacientes com sintomas leves da doença tem mais efeitos prejudiciais do que benéficos, reduzindo a resposta primaria do sistema imunológico contra a condição.
Por isso, cientistas começaram a procurar um composto capaz de controlar a tempestade de citocinas sem afetar o sistema imunológico – e o encontraram na natureza.

- Para criar o medicamento, os pesquisadores utilizaram um componente químico da Andrographis paniculata
- A planta é nativa da região da Índia, Sri Lanka e outras áreas do Sudeste Asiático e comumente utilizada com propósitos medicinais.

- O AG5 foi testado em larvas de peixe-zebra infectadas com um vírus e em camundongos com lesão pulmonar
- Em ambos os casos, o composto diminuiu consideravelmente os níveis de citocinas sem suprimir totalmente a resposta imunitária primária contra a inflamação.
- Especialmente no caso dos camundongos, houve redução de lesões pulmonares se comparado com os animais que não receberam tratamento com AG5.
- O composto não apresentou efeitos tóxicos em nenhum animal, mesmo quando administrado na dose máxima tolerada.
Aplicação em outras doenças inflamatórias
Segundo o New Atlas, os pesquisadores afirmam que o AG5 pode ser utilizado não só como tratamento alternativo nos casos de COVID-19, mas também para doenças inflamatórias crônicas, como artrite reumatoide, doença de Crohn, inflamação pulmonar e doença hepática gordurosa.
A equipe pretende realizar ensaio clínico para testar a segurança e eficácia do AG5 em pacientes com doença hepática gordurosa e deve fazer um pedido oficial de autorização à Agência Espanhola de Medicamentos e Dispositivos Médicos (AEMPS) ainda este ano.
